用厚玻璃制成，每块计数板由H形凹槽分为2个同样的计数池。计数池两侧各有一支持柱，将特殊的专用盖玻片覆盖其上，形成高0.10mm的计数池。计数池画有长、宽各3.0mm的方格，分为9个大方格，每个大格面积为1.0mm2.容积为0.1mm（ul），其中，大方格用双线分成25个中方格，位于正中及四角5个中方格是红细胞计数区域，用单线划分为16个小方格。四角的4个大方格是白细胞计数区域，用单线划分为16个中方格。根椐标准局（NBS）规定，大方格每边长度允许误差为±1%。
